The pandemic caused by the coronavirus COVID-19 has led to the widespread implementation of disinfection measures unprecedented in scale and an avalanche-like increase in the production of various types of disinfectants. Practice has shown that a high demand and lack of proper government control in many countries of the world, unfortunately, often leads to the cases when ineffective disinfectants are used.  Moreover, such cases are not always associated with low competence of business management, and in many cases have a corruptive nature and cause criminal consequences, especially when has a direct threat to the health and life of a large number of people.

The aim of our analysis is the study of some of the  peculiarities of Silver based disinfectants , which have recently begun to be widely used in various spheres of human activity. They have great prospects for application, namely, because of the use of the latest technologies for obtaining active substances of a new generation, which gives disinfectants other qualitative characteristics in comparison with other products in the market, that are alcohol and chlorine based,    compounds. Disinfection solutions, where silver is an active substance, can be divided into three groups.

The first group includes disinfectants containing silver salts. The most common of them is silver nitrate.

The second group includes disinfectants, where colloid silver is the active substance, as well as silver nanoparticles.

The third group includes disinfectants based on silver citrates.

The effectiveness of the use of all these drugs, as a rule, is enhanced by other active substances, such as, for example, hydrogen peroxide, alcohol, food acids.

Disinfectants of the first group, based on silver salts, including silver nitrate, have been known for a long time, but have not found widespread use due to their low efficiency. If they are mixed with hydrogen peroxide, then it performs the function of the main active substance. Thus, silver in nitrate form has low bioavailability and does not have a pronounced antibacterial and antiviral properties, that’s why it doesn’t show significant effects on basic functional properties of drugs.

Several Indian companies make disinfectants based on hydrogen peroxide and silver nitrate. The cost of the finished product is about $4/liter.

Nanotechnologies has opened wider possibilities for the production of new generation disinfectants using silver, and at present time there are several types of such disinfectants of this production method on the world market, which can be attributed to the second and third groups.

The second group of solution is obtained using outdated technologies of an unfinished cycle, where final product is the active substance in the form of silver nanoparticles. They can be used alone or in combination with other active ingredients. Most common of them is a combination of silver nanoparticles with hydrogen peroxide or alcohol.

Currently, the process of obtaining nanoparticles of silver or nanoparticles of any other metal is quite simple. However, the use of pure nanoparticles, including silver nanoparticles, is unsafe for humans and animals. This is due to the fact that nanomaterials have a biological effect and a complex of physical and chemical properties, which often radically differ from the properties of the same substance in the form of continuous phases or macroscopic dispersions (1).

  In the nanosized state, the following physicochemical features of nanoparticles behavior should be distinguished:

    1) an increase in the chemical potential of substances leads to significant changes in the solubility, reactive and catalytic ability of nanoparticles and their components;

       2) a large specific surface area of ​​nanoparticles increases their adsorption capacity, chemical reactivity and catalytic properties, leads to an increase in the production of free radicals and reactive oxygen forms, and the destruction of biological structures;

      3) small size and diversity of nanoparticles give them opportunity to bind to nucleic acids, proteins embedded in the membrane, penetrate the cell organelles and thereby alter the function of biological structures;

       4) high adsorption activity due to the developed surface of nanoparticles gives them the properties of an effective sorbent and the ability to absorb many times more substances per unit mass in comparison with macroscopic dispersions;

   5) high ability of nanoparticles to accumulation.

    It is clear that nanoparticles with the earlier mentioned physicochemical properties and the indicated biological action are completely new types of materials in comparison with traditional analogs. Therefore, silver nanoparticles can penetrate even through respirators into the human body who treats the room with such disinfectants. Also, there is a high possibility that remaining after processing on surfaces of solid interior space, they can also cause effects in accordance with the mechanisms described above in paragraphs 2, 3.

Thus, determination of potential risk to human health and life is required prior the application of disinfectants from the second group, where silver nanoparticles are active substances. 

A significant drawback of drugs of the second group is due to the peculiarities of the nanoparticles noted in paragraphs 1, 4, 5.

Thus, not stabilized silver nanoparticles come into uncontrolled reaction with any other molecules of aqueous medium or air, forming different insoluble compounds which tend to accumulation. In this case, the formation of large structures takes place, incommensurate with nano sizes, that have non-deterministic chemical composition. Therefore, due to the possibility of the formation of other compounds based on silver nanoparticles and the uneven distribution of silver as an active substance on the treated surface, its effectiveness as an antiviral component is significantly reduced. Below you can see photograph of a disinfectant in which silver nanoparticles are used as an active ingredient after 4 days of storage in daylight.

One can observe a change in color of the preparation and a significant size of macroscopic formations based on silver nanoparticles.

The low efficiency of drugs using silver nanoparticles was confirmed during comparative tests of disinfectants at the Institute of Epidemiology and Infectious Diseases named after V.L. Gromashevsky National Academy of Medical Sciences of Ukraine. The virucidal activity of existing disinfecting drugs on the Ukrainian market was investigated: "Sumerian Silver", "ISeS", "ARGUMIN", "BACTRILON A" (BAA-G), "Polidez" on the model of coronavirus of transmissible gastroenteritis of pigs (TGS).

For disinfecting drugs of the second group relates disinfectant «ARGUMIN», which is a colloidal solution of silver nanoparticles in demineralized water. 

A fragment of the report is presented below.

Determination of the effect of Solution "Sumerian Silver", "ISeS" , "ARGUMIN" , "BACTRILON A" (BAA-T), "Polidez" for extracellular coronavirus TGS was examined as follows: to a suspension of coronavirus TGS in a dose of 1000 TCD50/ ml was added solution "Sumerian Silver", "ISeS", "ARGUMIN", "BACTRILON A"  "Polidez" diluted 1:1000 were incubated at room temperature and were sampled in noncellular virus after 5 min and 20 min.

The results of determining the effect of drugs on extracellular coronavirus TGS are shown in Fig.2.

According to the obtained results, it was found that the disinfectants " Sumerian Silver", "BACTRILON A" (BAD-G), "Polidez" inhibited the reproduction of the extracellular coronavirus TGS by 2.0 lgID50 with an exposure of 30 minutes, and "ARGUMIN" only for 1.0 lgID50. A decrease in the infectious titer of the virus by 1.5-2.0 lgID50 and more in comparison with the control of the substance with the virus indicates the severity of the virucidal effect.

Thus, the disinfectant with silver nanoparticles does not have sufficient antiviral properties.

For this research was chosen one of silver nanoparticles disinfectants available on the market. Undoubtedly, similar results will be obtained for other disinfectants that use silver nanoparticles as active substances.

Unfortunately, many companies do not provide information on the chemical form in which silver is present in the preparation. For example, the American company “xxx” ™ is one example. Its composition is represented by the following active ingredients:

Hydrogen peroxide ………....5,00%

Silver ……………………… 0.01% 

Other ingredients ..................94.99% 

Total .. ……………………. 100.00%. 

However, there is no information on the chemical form of silver.

The third group of drugs should include silver citrate-based disinfectants. These are the most promising disinfectants with the following advantages:

- silver citrate does not have nanoparticles, has a high bioavailability, therefore it is completely safe for humans, can be used in their presence and used as an antiseptic;

- an aqueous solution of silver citrate is highly stable, which makes it possible to increase its shelf life up to 3 years;

- environmentally friendly, safe for people and animals by inhalation, ingestion, contact with mucous membranes;

- has no smell;

- does not cause allergies and dry skin when using drugs as antiseptics;

- has a prolonged effect.

The last four advantages make silver citrate disinfectants indispensable in comparison with alcohol and chlorine-containing preparations.

It is extremely important that when treating hands with disinfectants based on silver citrate, a person acquires and puts on a kind of “invisible antiviral silver gloves” that are effective for at least 24 hours. Then the chain of virus transmission from unwashed hands to the mucous membranes of the T-zone of a person (eyes, mouth, nose) breaks. A similar effect occurs in the processing of solid surface of interior in closed spaces, which are coated with invisible protective film and in case of virus contact with it, the virus is inactivated during all day.

Methods for producing silver citrate are among the most advanced innovative technologies. Therefore, in the world market there is a small amount of disinfectants of the third group.

In 2011, the company "Dupont" introduced to the world  market a drug “PURE Hard Surface” that was developed and patented by American company PURE Bioscience, Inc. , which has the following concentration of active ingredients:       

- silver: 0.003%;

- citric acid: 4.846%.

As was mentioned in a technical report (2), the drug inactivates the human coronavirus in 30 seconds. According to available information, such a disinfectant is sold on the American market for   not less than $21/liter and more.

Similar drugs with the same composition under the trademarks Axen®30 and Sdc3a developed by the same company PURE Bioscience, Inc. It was registered by the Agency for the Protection of Environment US EPA in the List N (3) as of 05.31.2020 year on the company ETI H2O Inc. Registration number is 729 77-3 (p.7) and 72977-5 (p.61) in the List N. The cost of these drugs is approximately not less than $20/liter.     Very interesting and new is the one described by PURE Bioscience, Inc. mechanism of action of silver citrate particles on viruses called "Trojan horse". The essence of the process is that the virus, like any other biological object, identifies any citrate, which is an organic compound, not as a foreign object that must be “feared”, but on the contrary, as a bioavailable substance that can potentially give impetus to development. Therefore, the virus very easily "captures" the citrate molecule, not distinguishing that there is a silver particle dangerous for its existence, which subsequently destroys the virus. Such a mechanism of inactivation of viruses is fundamentally impossible when using any other disinfectant available on the world market, including the preparations described above with silver of the first and second groups.   

We know that silver citrate disinfectants from PURE Bioscience, Inc. under various trademarks, except for the USA, are also used in Canada, Australia, Japan, Nigeria.   

In addition, such silver citrates are used as preservatives. For example, Silvérion 2400 is used at a concentration of 0.1 - 0.3% and is an approved preservative in the United States, and is currently included in the European list of approved preservatives. In Europe, Silvérion 2400 can also be used as an antimicrobial active agent. It is currently on the list of approved preservatives in Japan. This product complies with the 1986 California Safe Drinking Water and Toxicity Act because it is not established that it causes cancer, birth defects or other reproductive harm.     

The German company BASF has registered a preparation based on silver citrate for use in the cosmetic industry as a preservative.

However, at present, silver citrate has not yet been registered in Europe in accordance with (4, 5) as an active ingredient for disinfectants.

Thus, substances, including disinfectants using silver citrate are innovative and effective drugs of a new generation, with the help of which it is possible to trigger unique antiviral mechanisms, the presence of which cannot be imagined for any other disinfectants.

Similar preparations with the trademark "Sumerian Silver" were developed by the by our Team back in 2009 year , however, due to the monopolization of the national market of disinfectants, they are not widely used. The pandemic caused by the coronavirus COVID-19 has generated a rush of demand for disinfectants.

At present, NCare BioTech Inc. (NCare) in association with our partner company "Nanomaterials and Nanotechnologies" LLC,  produce the following solution using a patented technology.

Preparation «Sumerian Silver», which used as a disinfectant for premises and Hand Antiseptic. Its high efficiency as an antiviral drug has been confirmed by the Institute of Epidemiology and Infectious Diseases named after V.L. Gromashevsky of the National Academy of Medical Sciences of Ukraine in the study on the model of coronavirus, transmissible gastroenteritis of pigs (TGS) (6).   

The drug can be supplied as a concentrate with following composition:

Silver Citrate - 500 mg/L;

Copper Citrate - 500 m g/L; 

Purified water.

The concentrate must be diluted with demineralized water to 3% working solution, i.e. for 3 parts of the concentrate, add 97 parts of water.

The combined use of silver and copper gives a synergistic antiviral effect. However, if desired, the concentrate may be supplied without copper at a concentration of Silver Citrate - 500 mg/L. 

Also, a hand sanitizer under the trademark “Sumerian Silver” of the following composition can be supplied: 

Magnesium citrate - 35 mg/L;

Silver citrate - 35 mg/L;

Zinc citrate - 35 mg/L;

Copper citrate - 35 mg/L;

Purified water.

It can be concluded that the proposed solution “NCare”  in terms of the content of active substances are similar and not inferior to the conventional drugs obtained using the technology of the American company PURE Bioscience, Inc. Moreover, they are fortified with copper citrate as a synergistic component.  

But the main advantage of NCare "Sumerian Silver" preparations is that the unique technology of their production, which differs from the technology of the American company PURE Bioscience, Inc. It has a higher productivity that allows the production of active substances in very large volumes, while the prices for finished products are significantly lower than for similar products with silver citrate existing today on the world market.
